Betula pendula pollen, under laboratory conditions, was exposed to three atmospheric pollutants: carbon monoxide (CO), ozone (O3) and sulphur dioxide (SO2). Two levels of each pollutant were used; the first level corresponds to a concentration on the atmospheric hour-limit value acceptable for human health protection in Europe, the second level to a higher, at least more than double of the first, concentration level. Experiments were done under artificial solar light with controlled temperature and relative humidity. Our results indicate that, in urban areas, concentrations of CO, O3 and SO2 on the limits established for human protection, can affect pollen fertility. We verified a decrease in the viability and germination of the pollen, indicating damage to the pollen membrane system. Also, a general decreasing trend in the total protein content of the exposed samples when compared with the control samples was observed, which suggests alterations in the antigenic characteristics of pollen.  相似文献

The arthropod cuticle acts as a physiochemical barrier protecting the organism from pathogens' entry. Entomopathogenic fungi actively penetrate the cuticles of arthropod hosts and are therefore directly affected by cuticle composition. Previously we have observed that Metarhizium spp. developing on resistant ticks ultimately die without penetrating tick's cuticle, suggesting that the cuticles of resistant ticks have antifungal compounds. In the present study, lipids and water-soluble cuticular components were extracted from engorged female tick cuticles, of one susceptible and one resistant tick species to Metarhizium spp. While conidia exposed to lipids from the susceptible tick, Rhipicephalus annulatus, germinated and differentiated into appressorium, conidia exposed to lipids from the resistant tick, Hyalomma excavatum, were inhibited. Soluble cuticular component extracts from both susceptible and resistant ticks stimulated conidial germination but not appressorium differentiation. A comparative analysis of the fatty acid profile in lipid extract of each tick exhibited similar compositions, but the relative abundance of C16:0, C18:0, C18:1ω9C and C20:0 was 2–5 times higher in the extracts from resistant ticks. All of these fatty acids inhibited conidial germination in vitro at 1% and 0.1% w/v concentration, but C20:0 stimulated appressorium differentiation at low concentration. This is the first report demonstrating a possible link between the presence of antifungal compounds in a specific concentration in tick cuticle and tick resistance to infection.  相似文献

Seed germination and innate immunity both have significant effects on plant life spans because they control the plant's entry into the ecosystem and provide defenses against various external stresses, respectively. Much ecological evidence has shown that seeds with high vigor are generally more tolerant of various environmental stimuli in the field than those with low vigor. However, there is little genetic evidence linking germination and immunity in plants. Here, we show that the rice lectin receptor‐like kinase OslecRK contributes to both seed germination and plant innate immunity. We demonstrate that knocking down the OslecRK gene depresses the expression of α–amylase genes, reducing seed viability and thereby decreasing the rate of seed germination. Moreover, it also inhibits the expression of defense genes, and so reduces the resistance of rice plants to fungal and bacterial pathogens as well as herbivorous insects. Yeast two‐hybrid and co‐immunoprecipitation experiments revealed that OslecRK interacts with an actin‐depolymerizing factor (ADF) in vivo via its kinase domain. Moreover, the rice adf mutant exhibited a reduced seed germination rate due to the suppression of α–amylase gene expression. This mutant also exhibited depressed immune responses and reduced resistance to biotic stresses. Our results thus provide direct genetic evidence for a common physiological pathway connecting germination and immunity in plants. They also partially explain the common observation that high‐vigor seeds often perform well in the field. The dual effects of OslecRK may be indicative of progressive adaptive evolution in rice.  相似文献

Summary

We analyse the fruiting phase of the life cycleof Digitalis thapsi L. We studied the production and size of seeds and capsules, and the interpopulational variability and inter-relationships of these parameters. The capsule volume, number of seeds per capsule, of seeds per individual, and of capsules per individual were inter-related and fairly affected by the medium, whereas seed weight was found to bea more independent character. A germination study showed that the species has agreat germinative capacity and has positive photoblasty. It is non-dormant especially if incubated under experimental conditions close to the climatic conditions of the site of provenance of the samples. An increase in temperature speeds up the process without increasing germinability. The morphological and biometric characteristics of the first stages of development of the species following germination were analysed. It was found that seed weight is not positively correlated with cotyledon length nor with any of the other parameters con-sidered, and that the length of the fifth leaf is a useful parameter to express inter-populational genetic variability. Seedling production in spring seems to be influenced more by physical than by chemical factors.  相似文献

Background: Species persistence, particularly in monocarpic species, depends on the successful recruitment of individuals. An understanding of the factors that limit the recruitment of rare monocarpic plant species is therefore vital for their conservation.

Aims: To identify the factors limiting the recruitment of Rheum nobile, a rare and highly specialised monocarpic giant herb endemic to the high eastern Himalayas.

Methods: Seed sowing (seeds added or not added) and seedling transplanting experiments were conducted in disturbed (vegetation removed) and undisturbed plots in the vicinity of established populations of R. nobile to explore the mechanisms of recruitment limitation. Four levels of photosynthetically active radiation (0, 15, 30 and 50 μmol m?2 s?1) and two sowing positions (beneath and above grass litter or moss layer) were manipulated in the laboratory to determine how ground cover limited seedling emergence.

Results: Seed addition increased seedling recruitment. Disturbance significantly increased seedling emergence and establishment. Seed germination significantly decreased with the reduction of light availability, but 31.7% of all seeds germinated in complete darkness. Seedling emergence was close to zero when seeds were sown on top of a layer of grass litter or moss, but rose to 34.5% when the seeds were sown beneath such layers.

Conclusions: Our results indicate that the recruitment of R. nobile is limited by a combination of seed and microsite availability. Therefore, in order to conserve this species, we suggest adding seeds to suitable sites and implementing soil disturbances in existing populations to create suitable microsites.  相似文献

Abstract

Four different DNA extraction methods were used to extract genomic DNA of the medicinal mushroom Lingzhi from its developing stage materials, such as mycelium, dry fruiting body, or sliced and spore powder or sporoderm‐broken spore powder. The DNA samples were analyzed using agarose gel electrophoresis, UV spectrophotometer, and PCR amplification. According to the average yields and purity of DNA, high salt concentrations and low pH methods were the best for DNA extraction. The mycelia and sporoderm‐broken spore powder yielded higher and purer DNA. The method developed could effectively eliminate the influence of the secondary metabolites to DNA extraction. The DNA samples extracted from the developed method could be successfully used for PCR applications.  相似文献

Abstract

The effects of temperature and salinity (NaCl) on germination of Hordeum maritimum With, (halophyte) and H. murinum L. (glycophyte) seeds were investigated. Dehulled caryopses were used for monthly germination trials, starting from November (120 days of after‐ripening in darkness at 20±1°C). Trials were continued for one year. Differences in germination response between the two species were observed, confirming that H. Maritimum is better adapted to high salinity levels and to variations in external temperature than H. murinum. H. maritimum showed a germination control mechanism related to after‐ripening time and based on seed dormancy break/resumption. At higher temperature (30°C), thermodormancy was also recorded. No germination strategies were observed in H. murinum that is relatively insensitive to the combined effects of temperature and salinity. Thus, in virtually all treatments, H. murinum exhibited a higher germination rate compared with H. maritimum, as early as 72 h after imbibition, suggesting that dormancy, both in the presence or absence of salt, is totally abolished by early fall or at the latest in winter.  相似文献

Abstract

Changes in polyamine metabolism have been studied during early germination of Triticum durum Desf. cv. Cappelli. In the embryos of dry seeds, the adequate polyamine content decreases with a minimum at 36 h of water imbibition. A great need for polyamines during germination is expressed by reactivation of their biosynthetic enzymes. Putrescine biosynthesis mostly occurs via the ornithine–decar–boxylase pathway until 42 hours of hydration. Arginine–decarboxylase activity, almost absent in the first stages of imbibition, reaches its maximal level around 36–42 hours, when ornithine–decarboxylase falls. These changes suggest that the polyamine metabolism could be differently activated depending on the growth process related to the germination phases.  相似文献

Abstract

Germination tests were carried out on seeds from three Italian populations of Abies alba. Seed germinability was up to 38%. HgCl2, NaClO and plant preservative mixture were used as seed‐sterilising agents. Despite chemical treatments, Papulaspora rubida and Chaetomium globosum were isolated. The possible role of these fungi in the physiology of silver fir seed germination process was discussed.  相似文献
